You do not have to complete all these challenges in three rounds, so if you beat terrorists 3-1 and still have a challenge left is met, the match will continue until you have met all the requirements.

Leon pergi ke lokasi penculikan (sepertinya sich settingnya di Spanyol), ke sebuah desa bernama Puebos. Sesampai disana yg ia temukan adalah para penduduk yg kesurupan karena terinfeksi parasit (bukan virus). Parasit ini disebut Las Plagas dan manusia yg terjangkit dan telah pecah telur parasitnya disebut Las Ganados. Parasit ini diciptakan oleh Saddler, pemimpin aliran sesat di desa Puebos, Los Illuminados. Dlm pengembangan Parasit ini, Saddler menyewa seorg Scientist bernama Luis Sera. Namun Luis mengkhianati Saddler karena iya telah tau tujuan Saddler. Saddler sendiri memanfaatkan kepala desa Puebos, Chief Mendez dan Salazar. Salazar adalah pemimpin kerajaan di dekat desa Puebos, ia membantu Saddler karena ingin membalas hutang budi. Tujuan Saddler sendiri adalah menguasai dunia. Langkah pertamanya adalah menculi putri Pres utk dikendalikan utk mengambil alih Amerika. Dgn kekuatan Amerika, Saddler dgn mudah menguasai negara2 lain. Disaat itu, Wesker dr markasnya mengintainya. Dia tertarik dgn penemuan Las Plagas. Dia mengutus Ada Wong (What the!) dan Jack Krauser utk mengambil Samplenya. Ada sendiri memiliki tujuan sendiri begitu jg Krauser. Mrk bertiga saling tidak percaya. Dalam pencarian Ashley Graham (Putri Presiden), Leon bertemu Ada. Dlm permainan, Leon berhasil membunuh Chief Mendez dan Salazar. Seiring cerita, Leon dipertemukan oleh Krauser yg ternyata dialah yg menculik Ashley (jadi Krauser bekerja oleh Wesker dan Saddler jg). Dlm cerita, Krauser berhasil dikalahkan oleh Leon dan di finishing oleh Ada (Separate Ways). Saddler pun berhasil dikalahkan. Sample Las Plagas berhasil diambil Leon, namun Ada menodong Sample tersebut dan kabur. Ada ternyata telah men-set bom di pulau tersebut, Leon diberi kunci Motor-Boat utk melarikan diri bersama Ashley dan akhirnya kabur.
